description The taxonomic identification of species through analysis of the variation of orthologous DNA sequences, complement the information obtained with morphological characters. Cytogenetic studies indicate that polyploid taxa occur in the subfamily Opuntioideae, Opuntia ficus-indica, contributing to morphological variability in the individuals of a population, and influencing the correct identification of species. However, the lengths of the sequences in Opuntioideae are affected by the extraction of pure DNA. Different extraction methods were evaluated and modified, and a procedure was established to obtain good quality DNA, free of inhibitors for gene amplification by polymerase chain reaction. The ratio A260/A280 and A260/A230 ranged from 1.6 to 2.1, revealing absence of contamination with the modified protocol for DNA extraction from cotton leaves. This method is inexpensive compared to those of commercial manufacturers and, therefore, can be applied in studies with limited resources.

La identificación taxonómica de especies mediante análisis de variación de secuencias de ADN ortólogas, complementan la información obtenida con caracteres morfológicos. Estudios citogenéticos muestran taxones poliploides en la subfamilia Opuntioideae, Opuntia ficus-indica, contribuyendo a la variabilidad morfológica en los individuos de una población, e influyendo en la correcta identificación de las especies. Sin embargo, las longitudes de las secuencias en Opuntioideae están afectadas por la extracción de ADN puro. Se evaluaron y modificaron diferentes métodos de extracción y se estableció un procedimiento para obtener ADN de buena calidad, libre de inhibidores para amplificación de genes por reacción en cadena de la polimerasa. Las relaciones A260/A280 y A260/A230 variaron entre 1.6 a 2.1, revelando ausencia de contaminación con el protocolo modificado para la extracción de ADN a partir de hojas de algodón. Este método es de bajo costo comparado con los de casas comerciales y, por lo tanto, se puede aplicar en estudios con recursos limitados.
